Unsere Arbeitsgruppe untersucht Stammzellen, die aus dem dentalen Follikel isoliert werden können (DFVs). Der dentale Follikel ist ein Gewebe des Zahnkeims, aus dem der Zahnhalteapparat entsteht. Die DFVs besitzen ein Differenzierungspotential für Zellen des Zahnhaltapparats und insbesondere für biomineralisierende Zellen. Untersucht werden in unseren Projekten die molekularen Mechanismen von DFVs während der osteogenen Differenzierung, um die Bildung von Osteoblasten aus DFVs besser verstehen und steuern zu können. Dies ist eine wichtige Voraussetzung für eine Stammzellen-basierte Regeneration von Knochen.

Um DFVs in einer Zelltherapie zukünftig anwenden zu können, müssen diese ex vivo vermehrt werden. Aufgrund der zellulären Seneszenz, ist aber die Vermehrung somatischer Stammzellen begrenzt, was zu erheblichen Einschränkungen zukünftiger Zelltherapien mit DFVs führen kann. Wir untersuchen deshalb in unserer Arbeitsgruppe, welche exogenen Faktoren und zelluläre Signalwege an der Induktion der Seneszenz in DFVs beteiligt sind. Aus diesen Erkenntnissen wollen wir neue Zellkulturstrategien entwickeln, die die Seneszenz in DFVs unterdrückt.
